Semax is a synthetic heptapeptide (Met-Glu-His-Phe-Pro-Gly-Pro) built from the ACTH(4–7) fragment of adrenocorticotropic hormone, extended at the C-terminus with the tripeptide Pro-Gly-Pro. RESEARCH BACKGROUND
Work published in the Journal of Neurochemistry demonstrated that Semax binds specifically to membranes of the rat basal forebrain and raises levels of brain-derived neurotrophic factor protein in that region. Related studies reported regulation of BDNF and trkB expression in the hippocampus, placing the peptide within neurotrophic signalling rather than hormonal pathways.
Separate research has examined Semax in cerebral ischaemia models and reported activation of dopaminergic and serotonergic systems in rodents. First synthesised in Russia, it has been studied there for several decades and remains one of the most cited ACTH-fragment analogues in the neuropeptide literature.
